Soybean promoter activity is measured over the course of 100 h following introduction of soybean promoter constructs into lima bean cotyledonary tissue (Chiera et al, 2007). Promoter regions, recovered as the ~1.5 kb region upstream of the translational start, are first cloned into pFLEV (Finer Lab Expression Vector) containing the gfp gene. Following introduction via particle bombardment, GFP expression is tracked for 100 h using an automated image collection system (Buenrostro-Nava et al, 2006), and quantified to yield GFP expression profiles.
