Induction of Somatic Embryogenesis and shoots 

Murashige and Skoog salts
Gamborg B5 vitamins
3% sucrose

and either

1) 0.5 mg/l NAA, 5 mg/l BA

2) 0.1 mg/l 2,4-D

 FOR 1 LITER
10 ml    MS FeEDTA  add first
10 ml    MS Sulfates
10 ml    Halides
10 ml    P, B, Mo
10 ml    B5 Vitamins
1.65 g  NH4NO3
1.9 g    KNO3

volume up to 1000 ml

split into 2 x 500 ml

 

Shoots

250 µl (1 mg/ml) NAA
2.5 ml (1 mg/ml) BA

pH to 5.7

Embryos

0.25 ml (1 mg/ml) 2,4-D

pH to 5.7

 add 1.0 g Gelrite
autoclave, pour into plates

Students bring in any plant material of interest in an attempt to intiate cultures.
